Singh and a 19-member cabinet took the oath of office from Indian President Pratibha Patil at a ceremony at the presidential palace, in New Delhi on Friday.
His Congress party-led alliance was voted back to power, following marathon elections, which ended last week.
He is the only prime minister since the country's first post-independence leader Jawaharlal Nehru to be returned to office after completing a full five-year term.
Friday's swearing-in came amid a row between his Congress party and a key ally over cabinet posts.
